You should have the parking lights and flashers turned on. For many vehicles there is no need to have the engine running or ignition in the accessory position to light trailer lights when tow vehicle lights are lit on but I don't know about yours. There is no harm to have it running. Of course if you want to check a directional signal, then you need to have the tow vehicle powered up.
It is possible that your vehicle has a separate fused circuit for the trailer lights. If you get no voltage in the above test, you might want to find out if there is a separate fuse for the trailer lights.
